大麦单倍体的离体培养与快速繁殖

摘 要:

以大麦品种‘花30’小孢子培养来源单倍体植株作为起始材料, 分别对其丛生芽诱导、扩繁、生根以及再生植株水培等技术进行了研究。结果表明, 添加0.5 mg•L-1 噻苯隆(TDZ)的1/2MS培养基适宜丛生芽诱导; 添加1.0 mg•L-1 TDZ的1/2MS培养基适宜丛生芽增殖; 分别添加400 mg•L-1水解酪蛋白(CH)、20 mg•L-1亚精胺(Spd)和1.0 mg•L-1多效唑(MET)可以促进丛生芽的健壮; 添加0.05 mg•L-1 α-萘乙酸(NAA)和1.0 mg•L-1 MET的1/2MS培养基适宜生根。水培7~10 d后, 90%以上的再生植株正常生长。

Abstract:

In order to establish a stable barley haploid population, the haploid plants derived from microspore culture of barley (Hordeum vulgare) cultivar ‘Hua-30’ were used as materials, and the induction and rapid propagation of multiple buds, rooting and hydroponics of regenerated plantlets were studied. The results showed that the supplement of 0.5 mg•L-1 thidiazuron (TDZ) in 1/2MS basal medium was suitable for multiple buds induction; the supplement of 1.0 mg•L-1 TDZ in 1/2MS basal medium was suitable for bud rapid propagation and the addition of 400 mg•L-1 casein hydrolysate (CH), 20 mg•L-1 spermidine (Spd) and 1.0 mg•L-1 multieffect triazole (MET) improved the state of buds growth. The supplement of 0.05 mg•L-1 α-naphthaleneacetic acid (NAA) and 1.0 mg•L-1 MET in 1/2MS basal medium were suitable for rooting of regenerated plantlets, and more than 90% plantlets survived after an acclimation of 7–10 days by hydroponics.
